CJC-1295 vs Ipamorelin: Two Different Receptors
A GHRH analogue and a ghrelin receptor agonist, why the DAC distinction matters, and what the literature says about studying them together.
CJC-1295 and Ipamorelin are the two compounds most often requested together in growth-hormone research, and the reason is mechanical rather than promotional: they act on different receptors. This is what the published literature describes about each, and why they are studied as a pair.

Two different receptors
Growth hormone release from the pituitary is governed by two opposing inputs: GHRH, which stimulates it, and somatostatin, which suppresses it. Ghrelin acts on a third receptor and amplifies the response. The two compounds here occupy different points in that system.
CJC-1295 no DAC — also called modified GRF(1-29) — is a GHRH analogue. It is the first 29 amino acids of GHRH with four substitutions that resist enzymatic degradation, particularly at the DPP-4 cleavage site. The “no DAC” designation matters: the Drug Affinity Complex version binds albumin and extends the half-life to days, while the version without it is cleared in roughly half an hour. The literature treats these as different tools, not different brands.
Ipamorelin is a pentapeptide and a selective agonist of the growth hormone secretagogue receptor — the ghrelin receptor. Selectivity is the point of interest in the published work: earlier secretagogues in the same class also raised cortisol and prolactin, and studies describe Ipamorelin as producing GH release with markedly less effect on those axes.
Tesamorelin, for completeness
A third GHRH analogue appears in the same literature. Tesamorelin is a stabilised GHRH(1-44) analogue with a trans-3-hexenoyl group attached to the N-terminus, and the body of research around it is distinct — largely lipid-metabolism models rather than the pulsatility work that surrounds CJC-1295.

Why they are studied together
Because they act on separate receptors, the published rationale for combining them is that the effects are not redundant — one raises the stimulatory input, the other acts through a pathway that also suppresses somatostatin tone. Studies examining the pair describe a response greater than either alone.
This is a mechanistic argument, not a settled conclusion, and the combination literature is thinner than the literature on either compound individually. It is also worth noting that most of the combination work uses short-acting GHRH analogues specifically, which is why “no DAC” appears so often in these protocols.
Reading this literature critically
- Neither compound is an approved medicine in the European Union. Tesamorelin has regulatory approval in some jurisdictions for a specific indication; CJC-1295 and Ipamorelin do not.
- Much of the pulsatility work is in animal models, where the endogenous secretion pattern differs from the human one.
- Half-life figures vary considerably between studies depending on assay and species, so treat the numbers above as orientation rather than specification.
- The distinction between DAC and no-DAC versions is frequently blurred in secondary sources, which makes some published comparisons hard to interpret.
