SS-31 (elamipretide) is a synthetic mitochondria-targeted tetrapeptide with the sequence D-Arg-Dmt-Lys-Phe-NH2. Its aromatic-cationic structure lets it concentrate in the inner mitochondrial membrane independently of membrane potential.
Research centres on its selective binding to cardiolipin, a phospholipid of the inner mitochondrial membrane that is essential for electron-transport-chain function. Preclinical studies have described stabilised cytochrome c–cardiolipin interactions, more efficient electron transfer and reduced reactive-oxygen-species production in models of mitochondrial dysfunction, ageing and ischaemia–reperfusion.
