MOTS-c (Mitochondrial Open Reading Frame of the Twelve S rRNA type-c) is a mitochondrial-derived peptide (MDP) encoded within the 12S ribosomal RNA gene of mitochondrial DNA. Discovered in 2015 by Dr. Pinchas Cohen laboratory at USC, MOTS-c represents a novel class of signaling peptides that originate from the mitochondrial genome.

MOTS-c research has revealed its role as a key regulator of metabolic homeostasis. Studies have documented its effects on AMPK activation, folate-methionine cycle regulation, and cellular metabolism reprogramming. Remarkably, MOTS-c has been shown to translocate to the cell nucleus in response to metabolic stress, where it directly regulates gene expression — a process termed mitonuclear communication.

Often described in the literature as an exercise mimetic, MOTS-c has been reported in preclinical models to produce effects that parallel those of physical exercise — with changes in insulin sensitivity, fatty acid oxidation, and diet-induced weight gain described in rodent studies. Research has also investigated its decline with age and its potential role in the metabolic changes associated with aging.
